I received a traffic infringement notice for allegedly travelling 80km in a 40km zone at 12:03 am on the Western Ring Road. That's a $645 fine and 6 months drivers licence suspension. The part of the road is usually 100km zone. Do I have to take their word that it was a 40km zone? I've travelled that road for 10 years and have never received an infringement.

No you don't. You can elect to take it to court and make them prove their case. When their documentation arrives prior to the court date, you then go through it very carefully and see if there are any mistakes.

Pleading not guilty will entail at least 2 trips to court.

It will happen when it happens. Depends on what court, but think in months.

In the meantime you get to drive around.
